Title:Comparison of ELISA, dot-blot, and PCR assays for detection of whitefly-transmitted geminivirus

Abstract:The sensitivity and reliability of enzyme-linked immunosorbent assay (ELISA). dot-blot hybridization and polymerase chain reaction (PCR) were compared for detection of Abutilon mosaic geminivirus (AbMV). PCR assays were 10,000-fold more sensitive than ELISA and dot-blot hybridization assays for detection of AbMV. AbMV was detected by PCR from dilutions of leaf samples equivalent to 10 ng infected leaf tissue. In Hawaii, the sweet potato whitefly B. tabaci biotype B causing silverleaf and other disorders on vegetable crops. The establishment of three diagnostic assays for detection of WFT geminiviruscs in Hawaii will enable us to monitor any new occurrences of WFT geminiviruses, and to study epidemics of these new WFT geminiviruses in Hawaii.
